Reconstituting a peptide means dissolving a freeze-dried (lyophilized) amount in a vial with a liquid diluent, usually bacteriostatic water. Once it's dissolved, the whole calculation is just three steps:
- Concentration = vial amount ÷ diluent volume
- Dose volume = prescribed dose ÷ concentration
- Syringe units = dose volume × the syringe's units-per-mL
Worked example
Say you've reconstituted a 10 mg vial with 2 mL of bacteriostatic water, and you've been prescribed a 250 mcg dose, using a U-100 insulin syringe (100 units per mL):
- Concentration: 10 mg ÷ 2 mL = 5 mg/mL
- Convert the dose to match: 250 mcg = 0.25 mg
- Dose volume: 0.25 mg ÷ 5 mg/mL = 0.05 mL
- Syringe units: 0.05 mL × 100 = 5 units
So you'd draw to the 5-unit mark on a U-100 syringe.
