The three numbers reconstitution comes down to

Reconstituting a peptide means dissolving a freeze-dried (lyophilized) amount in a vial with a liquid diluent, usually bacteriostatic water. Once it's dissolved, the whole calculation is just three steps:

  1. Concentration = vial amount ÷ diluent volume
  2. Dose volume = prescribed dose ÷ concentration
  3. Syringe units = dose volume × the syringe's units-per-mL

Worked example

Say you've reconstituted a 10 mg vial with 2 mL of bacteriostatic water, and you've been prescribed a 250 mcg dose, using a U-100 insulin syringe (100 units per mL):

  1. Concentration: 10 mg ÷ 2 mL = 5 mg/mL
  2. Convert the dose to match: 250 mcg = 0.25 mg
  3. Dose volume: 0.25 mg ÷ 5 mg/mL = 0.05 mL
  4. Syringe units: 0.05 mL × 100 = 5 units

So you'd draw to the 5-unit mark on a U-100 syringe.
